چکیده
The crude latex extract of Asclepias curassavica contains many proteins and exhibits strong proteolytic activity. The study was done to identify the clotting factor present in the latex of Asclepias curassavica and to analyze the thrombin like activity of the clotting factor. In this study the plant latex from Asclepias curassavica was collected and extracted the proteins. Proteolytic assays and inhibitor assays were carried out in the crude sample extract. The docking studies were carried out using Bioinformatics tools. The plasma clotting time was reduced from 143 seconds to 56 seconds in the presence of the crude latex extract. Reduction in coagulation time was observed with increase in the concentration of crude extract. In-silico studies showed that the cysteine protease of Asclepias curassavica interacts with the coagulation factors. The Factors VIII and XI there by leading to clot formation. Hence, from this research, it was proved that the compounds, Asclepain cI and cII, were readily involved in the clotting activity and were also found to reduce the time of coagulation.

Microsciadin, a New Milk-Clotting Cysteine Protease from an Endemic Species, Euphorbia microsciadia
In the present work, a new branch of biotechnological advantage of the latex of an endemic perennial plant, Euphorbia microsciadia has been introduced. A novel cysteine protease, designated as microsciadin, was purified from the latex of Euphorbia microsciadia by a combination of sequential usage of SP-Sepharose Fast Flow column in two different pHs and a final gel filtration ...
متن کاملBiochemical analysis of a papain-like protease isolated from the latex of Asclepias curassavica L.
Most of the species belonging to Asclepiadaceae family usually secrete an endogenous milk-like fluid in a network of laticifer cells in which sub-cellular organelles intensively synthesize proteins and secondary metabolites. A new papain-like endopeptidase (asclepain c-II) has been isolated and characterized from the latex extracted from petioles of Asclepias curassavica L. (Asclepiadaceae). As...
متن کاملMacroevolutionary Trends in the Defense of Milkweeds against Monarchs Latex, Cardenolides, and Tolerance of Herbivory
Theory predicts that plants will increase their defenses against herbivores over time; however, several milkweed defenses have declined as the genus Asclepias has diversified. We review the evolutionary history of milkweed defense and provide new findings from an experiment on 53 Asclepias species. As predicted, monarch performance increased on more recently evolved species, and latex was prima...
متن کاملCorneal Toxicity Following Exposure to Asclepias Tuberosa
PURPOSE To present a case of corneal toxicity following exposure to milky plant latex from Asclepias tuberosa. METHODS A 70-year-old female presented with blurred vision and pain in her left eye after handling an Ascepias tuberosa. Clinical examination revealed a corneal stromal oedema with small epithelial defects. The corneal endothelium was intact and folds in Descemets membrane were obser...
متن کاملEvolution of latex and its constituent defensive chemistry in milkweeds ( Asclepias ) : a phylogenetic test of plant defense escalation
A tremendous diversity of plants exude sticky and toxic latex upon tissue damage, and its production has been widely studied as a defensive adaptation against insect herbivores. Here, we address variation in latex production and its constituent chemical properties (cardenolides and cysteine proteases) in 53 milkweeds [ Asclepias spp. (Apocynaceae)], employing a phylogenetic approach to test mac...
